Pringsheim's Soil Media

This is good for photosynthetic protists with chlorophylls a and c (yellow brown in color).

1. Put 1 grain of calcium carbonate in the bottom of each of 10 culture tubes Cover the CaCO3 with one inch of sterile soil.

2. With the squirt bottle of spring water, moisten the soil in each tube.

3. Allow the culture tube to sit for about one minute.

4. Gently add two inches of spring water to each tube (The end result should not be muddy water! You want about 2 inches of clear water sitting on top of the soil).
